The Faculty of Medicine, UKM was established in 1974 and began its academic session with the intake of the first group of 44 students for the 1973/74 intake session. Originally, the main campus was located on Jalan Pantai Baru Lembah Pantai and was later moved to the Wooden Building in the Kuala Lumpur Hospital Complex (HKL).

Originally, the main campus was located on Jalan Pantai Baru Lembah Pantai and was later moved to the Wooden Building in the Kuala Lumpur Hospital Complex (HKL). On 27 February 1975, HKL was declared as the Teaching Hospital of the UKM Faculty of Medicine. The permanent building of the Faculty of Medicine was built next to HKL in 1977. In 1979, UKM successfully produced the first group of UKM Medical Doctors. UKM has established its own teaching hospital, HUKM, which was established on 02 November 1993. Construction of HUKM began on a 23-hectare site in Jalan Yaakob Latiff, Bandar Tun Razak, Cheras Kuala Lumpur. On 01 July 1997, HUKM was completed and began operations equipped with various modern and latest equipment and was officiated by YAB Tun Dr. Mahathir Mohamad, the then Prime Minister of Malaysia on 14 July 1998.
